黄芪注射液对IgA肾病患者免疫球蛋白分子异常糖基化及Cosmc基因表达的影响

摘要：

目的探讨黄芪注射液对IgA肾病患者免疫球蛋白分子异常糖基化及Cosmc基因表达的影响。方法选择Ig A肾病患者26例,分离患者外周血B淋巴细胞并分别培养于RPMI1640培养基(空白对照组)、RPMI1640+黄芪注射液200 mg/ml(黄芪低剂量组)、RPMI1640+黄芪注射液1 000 mg/ml(黄芪中剂量组)和RPMI1640+黄芪注射液2 000 mg/ml(黄芪高剂量组)。经培养72 h后检测上述4组细胞中Cosmc基因mRNA及蛋白表达水平,酶联免疫吸附(ELISA)法检测上清液中IgA1水平。结果与空白对照组比较,加入黄芪注射液后B淋巴细胞中Cosmc基因mRNA及蛋白表达水平显著升高(P<0.05),且黄芪高剂量组显著高于中、低剂量组(P<0.05);与空白对照组比较,加入黄芪后B淋巴细胞中培养基上清IgA1水平显著降低(P<0.05),且黄芪高剂量组显著低于中、低剂量组(P<0.05);与空白对照组比较,黄芪注射液组IgA1与蚕豆凝集素结合力明显下降(P<0.05),且高剂量组下降更为显著(P<0.05)。结论黄芪可能通过上调Cosmc基因表达水平调控IgA肾病患者免疫球蛋白分子IgA1异常糖基化。
