葫芦素E通过诱导自噬抑制人膀胱癌细胞T24的增殖

摘要
目的探讨葫芦素E(CuE)对人膀胱癌细胞T24自噬的影响,并初步研究所诱导的自噬对细胞增殖的影响。方法通过MTT试验检测CuE对T24细胞增殖的影响。细胞经CuE处理后,通过western-blot检测自噬相关蛋白LC3A/B和p62的表达。通过LC3翻转试验和GFP-RFP-LC3双荧光试验检测自噬流的发生。Western-blot检测自噬抑制剂3-MA对CuE所诱导自噬的抑制作用,MTT试验和细胞计数试验研究3-MA抑制自噬后CuE对细胞增殖的影响。结果 CuE抑制T24的增殖,24 h的IC50约为0.75μmol/L。T24细胞经CuE处理后,LC3A/B II表达及LC3A/B II/I比值增高(P <0.05)、自噬经典底物蛋白p62表达减少(P <0.05)、LC3翻转试验阳性(P <0.05)以及LC3双荧光试验黄色和红色荧光颗粒的数目明显增多(P <0.05),提示CuE诱导了自噬的发生。3-MA能够抑制CuE所诱导自噬的发生(P <0.05),3-MA抑制自噬后,CuE对T24的增殖抑制效应减弱(P <0.05)。结论 CuE在人膀胱癌细胞T24中诱导了自噬的发生,且诱导的自噬对细胞的增殖起抑制作用。
