当归芍药散对快速老化模型小鼠海马蛋白表达的影响

摘要
目的研究当归芍药散(powder of Chinese Angelica and Peony,DSS)对快速老化模型小鼠(senescence-accel-erated mouse,SAM)海马蛋白表达的影响。方法选择12月龄SAM的快速老化亚系SAMP8和抗快速老化亚系SAMR1作为研究对象,分为SAMR1组、SAMP8组和SAMP8+DSS组(3.2g/kg),每组10只,连续给药30d。应用双向凝胶电泳结合质谱鉴定,观察DSS对SAMP8海马蛋白表达的影响。结果与SAMR1组比较,SAMP8组海马中有9个蛋白表达下调,13个蛋白表达上调;SAMP8+DSS组与SAMP8组比较,海马中有14个蛋白表达下调,21个蛋白表达上调。经肽质量指纹谱分析及蛋白数据库检索,表达有明显变化的蛋白根据功能可分为5类:线粒体相关蛋白、能量代谢相关蛋白、突触形成相关蛋白、细胞骨架蛋白、其他蛋白。结论SAMP8和SAMR1海马蛋白表达存在明显差异,DSS可影响快速老化模型小鼠海马蛋白的表达,其作用是多途径多靶点的。
