局灶性脑缺血大鼠VEGF/Notch1信号分子的表达及脑泰方的调节作用

摘要
目的从VEGF-Notch1信号通路观察脑泰方对局灶性脑缺血大鼠的治疗性血管新生样作用。方法将65只SD大鼠随机分为假手术组、MCAO组(模型组)、脑泰方组(NTF)、NTF+ED(重组人血管内皮抑制素)组。线栓法制备大鼠大脑中动脉栓塞(MCAO)模型,NTF组及NTF+ED组分别于造模成功后24 h灌胃NTF或灌胃NTF加腹腔注射ED,术后1、7、14、28 d取脑,采用RT-PCR、Western印迹方法观察血管内皮样生长因子(VEGF)、Flk-1、Notch1的表达。结果与MCAO组比较,NTF组大鼠各时间点缺血侧脑组织VEGF、Flk-1、Notch1表达水平升高,在第14天时达高峰,而加用血管内皮生成抑制素恩度后,这种增高效应随即消失。结论脑泰方可能通过上调VEGF-Notch1信号通路的表达以达到治疗性血管新生样作用。
