西红花苷在脑卒中后抑郁大鼠中抗炎作用的研究

摘要
目的探讨西红花苷对脑卒中后抑郁(post-stroke depression,PSD)大鼠的保护作用,并初步探讨其可能的作用机制。方法选用Wistar大鼠行卒中术后7天给予慢性不可预见性刺激结合孤养21天制作PSD模型。60只健康成年雄性Wistar大鼠,随机分为4组:卒中组、PSD组、PSD+西红花苷组和PSD+IL-1拮抗剂组。饲养30天后,分两方面进行检测:行为学方面行敞箱实验和糖水消耗实验检测;炎症指标利用Western blot法检测炎症因子IL-1的表达。结果行为学方面,与卒中组大鼠比较,PSD组大鼠糖水消耗量和敞箱实验中穿越格子数及站立次数明显减少(P<0.05);与PSD组大鼠比较,PSD+西红花苷组和PSD+IL-1拮抗剂组大鼠均能显著增加糖水消耗量和敞箱实验中穿越格子数及站立次数(P<0.05),但该两组间比较差异无统计学意义。炎症因子方面,与卒中组大鼠比较,PSD组大鼠脑组织中IL-1的表达有明显增加,差异有统计学意义(P<0.05);与PSD组大鼠比较,PSD+西红花苷组和PSD+IL-1拮抗剂组大鼠脑组织中IL-1的表达均显著降低,但该两组间差异无统计学意义。结论西红花苷可改善PSD大鼠的抑郁状态,其机制可能是通过抗炎作用实现的。
