摘要：

目的 :简化Northern印迹的操作步骤 ,建立安全无毒的RNA凝胶电泳方法。方法 :RNA经变性处理后在不含任何化学物质的中性琼脂糖凝胶上电泳 ,然后用 0 0 5mol LNaOH溶液转印 3h至转印膜上 ,相继与特异核酸探针杂交 ,检测不同表达丰度的基因 ,放射自显影成像。结果 :凝胶电泳图显示清晰的 2 8S和 1 8SrRNA条带 ;Nothern杂交检测出低丰度表达基因。结论 :改良的Northern印迹法能够达到传统方法有效变性、分离、检测特异基因表达的效果 ,垂直凝胶电泳可检测低至 5ng的RNA。
