桃仁膝康丸激活软骨细胞自噬延缓骨性关节炎软骨退变的研究

摘要
目的观察桃仁膝康丸(TRXK)对外科手术诱导的大鼠骨性关节炎(OA)软骨退变的保护作用并探讨其与软骨细胞自噬的关系。方法将健康雄性SD大鼠40只随机分为:假手术组、模型组、TRXK组和TRXK+自噬抑制剂3-甲基腺嘌呤(3-MA)组,采用改良的Hulth法制备大鼠膝关节OA模型。术后2周开始TRXK组和TRXK+3-MA组给予TRXK 1.25 g·kg-1·d-1灌胃,TRXK+3-MA组同时给予3-MA 1.5 mg·kg-1·d-1腹腔注射,连续用药12周。光镜下观察软骨组织形态学变化,并进行Mankin评分,Real-time PCR法检测膝关节软骨组织中Ⅱ型胶原α1(COL2A1)、聚集蛋白聚糖(Aggrecan)、基质金属蛋白酶13(MMP13)和血小板反应蛋白解整合素金属肽酶5(ADAMTS5)m RNA的表达,Western Blot法检测软骨组织中ULK1、Beclin-1和微管相关蛋白1轻链3-Ⅰ/Ⅱ(LC3-Ⅰ/Ⅱ)蛋白质的表达。结果与模型组比较,TRXK能明显降低Mankin评分,升高软骨组织中COL2A1和Aggrecan,而降低MMP13和ADAMTS5 m RNA表达(P<0.05);另外,TRXK能提高ULK1、Beclin-1和LC3-Ⅱ蛋白质表达(P<0.05)。结论桃仁膝康丸可以通过诱导软骨细胞自噬延缓或阻止外科手术诱导的大鼠膝关节OA的软骨退变,其激活软骨细胞自噬的机制有待进一步研究。
