Kinetics of desorption of proteins from the surface of protein-coated alumina by various desorbing reagents

摘要
The phenomena of desorption of adsorbed proteins have been studied in detail, The extents and rates of desorption of lysozyme from the surface of a protein-coated alumina by different desorbing reagents have been studied, The extent and rate of desorption of lysozyme preadsorbed at pH 7.0 are found to be maximum in the presence of 2 M NaCl solution and in the presence of a buffer of pH 5.0, The rate of desorption of three proteins, BSA, gelatin, and beta-lactoglobulin, preadsorbed at pH 5.0 on the surface of alumina by a buffer of pH 7.0, were also studied, As in the case of adsorption, the kinetic data fit into a first-order equation with two rate constants K-d(1) and K-d(2). The effect of temperature on the rate of desorption of BSA and lysozyme has been studied. The various activation energy parameters for the desorption of proteins have been calculated and a mechanism of protein desorption has been proposed. (C) 1996 Academic Press, Inc.
