Use of calcein and SYTO-13 to assess cell cycle phases and osmotic shock effects on E-coli and Staphylococcus aureus by flow cytometry

The fluorochromes SYTO-13 and calcein were tested by flow cytometry for their ability to assess the cell phases and the effects of osmotic shock in E. coli and S. aureus after exposure to NaCl and sucrose. Results were validated by viable plate count. Flow cytometry gave quicker results and more accurate information about intermediate states and population heterogeneity of S. aureus and E. coli than viable plate counts. SYTO-13 was an excellent marker for total counts and calcein can be used to assess esterase activity in S. aureus. (C) 1999 Elsevier Science B.V. All rights reserved.
