Direct bioelectrochemical monitoring of choline oxidase kinetic behaviour in Langmuir-Blodgett nanostructures

摘要
This study focuses on the potentiality of a direct interfacing of an enzyme-langmuir-Blodgett (LB) layered structure with a chemical transducer, where recognition and transduction are conducted in a single device. Choline oxidase (ChOD) was inserted in an hydrophillic or hydrophobic environment of behenic acid (BA) LB films and the biosensing layer was directly coated on a specially designed amperometric transducer. The observed Michaelis constant of choline oxidase for choline was determined and the results showed that the sensor response directly reflected the enzyme kinetics. This application of the biosensor concept enables us to investigate the immobilized enzyme kinetic behaviour in a biomimetic situation which involves a proteo-lipidic layered nanostructure.
