Catalysis of hydrolysis of a phosphate diester by mono- and dinuclear macrocyclic zinc(II) complexes

The macrocyclic ligand 3,6,9,17,20,23-hexaazatricyclo[23.3.1.1(11.15)]1, 11, 13, 15, 25, 27-hexaene (BMXD) forms a variety of mononuclear and dinuclear zinc complexes in 75% ethanol. Monohydroxo complexes of both the mononuclear and dinuclear species form in solution. The ability of these monohydroxo complexes to catalyze the hydrolysis of bis(4-nitrophenyl) phosphate (BNP) was studied. The L-Zn-OH complex forms at a higher pH than does the dinuclear analog, and is more effective at hydrolyzing BNP than L-Zn-2-OH. Mechanisms for the hydrolysis of BNP are proposed, exploring the coordination mode of the substrate and the nucleophilicity of the attacking hydroxide. (C) 1999 Elsevier Science S.A.
