Single-run determination of adenylate nucleotides, and of cellular energy status, by a simple and improved capillary electrophoretic method

摘要
A sensitive, convenient and reliable free-zone capillary electrophoretic method has been developed for the determination of adenylate nucleoticles in cell lysates without pre-analytical sample clean up. Simultaneous quantification of AMP, ADP and ATP can be performed with precision (within run) of 1.7 - 2.7% and reproducibility (day to day) of 1.3 - 3.5%. Detection limits for AMP, ADP, and ATP were 10 pm each and the linear range was 10 - 200 muM for each. The sample volume required was less than 10 nL. The proposed method greatly simplifies and accelerates determination of cellular 'adenylate energy charge', the most reliable measure of cellular energy status.
