Cloning of a single-copy DNA sequence unique to Plasmodiophora brassicae

A 1.4 kb DNA fragment was amplified from Plasmodiophora brassicae by the polymerase chain reaction using a primer PBTZS-2 (5'-CCGAATTCGCGTCAGCGTGA-3'). The 1.4 kb fragment was found in all isolates examined in this study. Southern blot analysis showed that the DNA sequence existed as a single copy per haploid genome of P. brassicae. The 1.4 kb fragment was cloned and sequenced. Database searches did not reveal any significant homologies with previously characterized nucleotide sequences. A nested primer pair PBTZS-3 (5'-CCACGTCGATCACGTTGCAAT-3') and PBTZS-4 (5'-GCTGGCGTTGATGTACTGGAATT-3') derived from the fragment produced a single 398 bp product from ail isolates of P. brassicae and no products from other fungi and bacteria examined. The potential use of the primers (PBTZS-2, PBTZS-3, and PBTZS-4) for specific detection of P. brassicae is discussed. A method for preparation of high molecular weight DNA from lyophilized resting spores of P. brassicae is also described. (C) 1997 Academic Press Limited.
