摘要
A method for determining K(m) of mushroom tyrosinase as well as the maximum velocity using L-dopa and L-tyrosine as substrates is described. The technique is based on measurements of the oxidation current of L-dopa which is both the substrate for the catecholase activity and the product of the enzymatic oxidation Of L-tyrosine. The oxygen concentration in the solutions is kept constant al the saturation level with air. A vitreous carbon electrode activated in 1 M KOH is employed. The values of the catalytic parameters obtained here with the voltammetric technique are comparable with those obtained by other authors under similar operative conditions using the spectroscopic method.
