DETERMINATION OF ENANTIOMERIC EXCESS BY FLUORESCENCE-DETECTED CIRCULAR-DICHROISM

The first determination of enantiomeric excess using fluorescence-detected circular dichroism (FDCD) without physical separation of the enantiomers is demonstrated for mixtures of the binaphthyl compounds (S)- and (R)-1,1'-binaphthyl-2,2' diyl hydrogen phosphate(BNPA). The FDCD measurement provides excellent accuracy in the determination of enantiomeric excess as well as excellent detectability of the enantiomeric compound, with a detection limit of 20 nM, corresponding to circular dichroism signal Delta A of 3.4 x 10(-7) absorbance units, for pure (S)-BNPA. Unlike conventional techniques that require two detectors, only one detector is necessary for the FDCD determination. The enantiomeric excess is shown to be equal to the FDCD signal divided by the Kuhn dissymmetry factor of the enantiomeric compound. Since the dissymmetry factor is independent of the concentration of the compound, enantiomeric purity can be determined for any total concentration of the compound within its linear dynamic range, once the dissymmetry factor has been determined. The technique is applicable to fluorescent enantiomeric compounds that exhibit FDCD activity.
