A comparison was made between high performance liquid chromatography (h. p. l. c. ) and colorimeteric and gas chromatography (g. c. ) methods for quantitating the conversion of wood derived sugars to a variety of solvents. Steam-treated aspenwood, which was previously pretreated to selectively remove the lignin or hemicellulose components, was subsequently extracted into a xylan rich water-soluble fraction and a cellulose rich water-insoluble fraction. The hydrolyzed fractions were used as the substrates for growth and solvent production by either Klebsiella pneumoniae, Clostridium acetobutylicum and Candida shehatae. Sugar utilization and solvent production could be determined by a single h. p. l. c. analysis and gave values that were comparable to those determined by g. c and colorimetric methods.
