FLOW-INJECTION SOLID-PHASE CHEMILUMINESCENT IMMUNOASSAY USING A MEMBRANE-BASED REACTOR

摘要
A flow injection analysis (FIA) sandwich immunoassay (ELISA) using a polyvinylidene difluoride polymer based membrane immunosorbent has been proposed. The membrane to which the antigen (bovine IgG) is attached is mounted in a flow cell (5-mu-L dead volume). Analyte in the form of mouse antibovine IgG is injected into the flowing stream followed by a goat anti-mouse IgG horseradish peroxidase (HRP) conjugate. The HRP was used to catalyze the enhanced luminol reaction, resulting in chemiluminescence, which was detected directly within the immunoreactor. A detection limit of 1 fmol was obtained while spiked serum samples gave precision of 8.7%.
