1. A survey is made of the induction of auxotrophy in Aerobacter (Klebsiella) aerogenes by MNNG (N-methyl-N′-nitro-N-nitrosoguanidien). 2. Of a variety of simple media of pH 6.0, the yield of auxotrophs was highest on treatment in tris-maleic acid buffer but, in general, the lethal action increased the more closely the medium approached to a complete (minimal) growth medium. In nutrient broth (pH 7.5) the low activity of MNNG was potentiated by the addition of nucleotide bases; adenine, guanine and cytosine increased mutagenesis considerably, and hypoxanthine the lethality. 3. Possible mechanisms for the observed effects are discussed. © 1969 Springer-Verlag.
