The gene and cDNA (TUB1) encoding an alpha-tubulin (alpha-Tub) from Pneumocystis carinii carried in rats were isolated and sequenced. The gene produced a 1.4-kb transcript that contained an open reading frame of 1347 nucleotides coding for a protein of 50 326 Da. The nt sequence of TUBI of P. carinii was 65-71% identical to other TUB1 genes, including those of yeast and human. The deduced amino acid sequence of P. carinii alpha-Tub showed 75-84% identity to other alpha-Tubs. Eight small introns resided within TUB1. P. carinii TUB1 was mapped to a 425-kb chromosome. The cloned gene appears to be the only alpha-Tub-encoding gene in the P. carinii genome.
