LASER-INDUCED FLUORESCENCE DETECTION IN CAPILLARY ELECTROPHORESIS WITH BLUE-LIGHT FROM A FREQUENCY-DOUBLED DIODE-LASER

A compact fluorescence detector has been developed for capillary electrophoresis (CE) based on a stabilized frequency-doubled semiconductor diode laser. By using a quasi-phase matching waveguide for frequency doubling, it was possible to obtain blue light with a wavelength of 424 nm, which is close to one of the excitation maxima of naphthalene-2,3-dicarboxaldehyde (NDA). Using an epiluminescence setup, a detection limit of 0.9 amol was obtained. It is shown that the laser power of 0.5 mW generated in this system is higher than needed for the best performance. Applications of CE separation of amino acids and body fluids are shown.
