PULMONARY DRUG-DELIVERY OF FINE AEROSOL-PARTICLES FROM SUPERCRITICAL FLUIDS

摘要
Better methods for delivery of pharmacologic agents directly to and via the lung are needed. We found that a solution of alpha-tocopherol (vitamin E) dissolved in supercritical carbon dioxide formed 0.3 to 3 mum diameter droplets when the solution was allowed to expand through a restrictor nozzle. The decompressed aerosol stream was diluted with gaseous oxygen and directly administered to rats for 10 to 30 min via inhalation. These treatments resulted in significant increases in the lung vitamin E levels of the rats. The supercritical fluid drug delivery (SFDD) method holds potential for pulmonary delivery of many other drugs.
