陕北滩枣多糖的单糖组成分析及其药理功效研究

摘要
本研究旨在描述陕西滩枣多糖(Zizyphus jujube cv. Shaanbeitanzao, ZSP)的单糖组成,并且探究滩枣多糖抗氧化、抗急性肝损、预防胰岛素耐受、预防血脂异常等药理功效。高效液相色谱分析表明,ZSP的主要单糖为L-阿拉伯糖(50.2%,摩尔百分比)。除了表现出强大的体外抗氧化活性外,其抗氧化活性通过CCl4诱导的急性肝损伤小鼠得以进一步验证(100,200, and400mg/kg·BW)。结果证实,ZSP (400mg/kg·BW)不仅显著降低了小鼠血清谷丙转氨酶(ALT).谷草转氨酶(AST)、乳酸脱氢酶(LDH)在血清以及小鼠肝组织丙二醛(MDA)水平的上升(p<0.01)。接受ZSP预处理的小鼠(400mg/kg·BW),其肝脏指数(HI),以及肝组织谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)均得以有效控制(p<0.01)。此外,连续饲喂20%果糖饮用水4周的小鼠,表现出高血糖、高胰岛素、血脂异常以及胰岛素敏感性受损等病理症状(p<0.05)。而口服400mg/kg·BW剂量的滩枣多糖,不仅能够使血清葡萄糖、胰岛素和血脂水平恢复正常,还可以明显改善动物胰岛素抵抗水平(HOMA-IR)和胰岛β细胞功能(HOMA-β)。 结果表明,ZSP不仅通过自身抗氧化与自由基清除功效发挥拮抗CCl4诱导的小鼠急性肝损伤,而且能够显著地改善高果糖饮水导致的胰岛素抵抗和血脂异常等小鼠症状。
