中药乳香的化学成分及生物活性研究

摘要
天然产物研究为药物研发提供结构新、疗效好、毒性低的活性物质,成为新药研究的先导和源泉。本文选用的天然产物为橄榄科的乳香(Boswellia carterii),对其化学成分分离、纯化、结构确证与乳香酸系列化合物的制备及抗肿瘤活性进行研究,具体分为以下3部分: 1.乳香系安徽毫州国芍堂中药科技有限公司提供,产地为索马里,将乳香粉碎,99%乙醇提取得浸膏,用乙酸乙酯、正丁醇分别与水进行萃取分配。各部分采用常规的硅胶柱层析、制备薄层层析、反相柱层析、凝胶Sephadex LH-20及重结晶等手段分离到17个化合物,采用现代波谱技术(UV、EI-MS、1H-NMR、13C-NMR、DEPT、1H-1H COSY、HMQC、HMQB和NOESY等),确定了大环二萜如Incensole,四环三萜如KTDA,五环三萜如AKBA等共14个化合物的结构。 2.乳香中含有天然的抗肿瘤化学成分3-羰基甘遂-8,24-二烯-21-羧酸(KTDA)与3-乙酰氧基-11-羰基-β-乳香酸(AKBA),为提高其抗肿瘤活性,在总结先前关于KTDA以及乳香酸类似物抗肿瘤活性研究文献的基础上,将KTDA经酯化和胺化得到7个KTDA衍生物,同样方法得到3个AKBA衍生物。 3.采用MTT法,将乳香中分离出的化合物7(新化合物)以及KTDA和AKBA的系列衍生物进行体外抗肿瘤活性测试,选用了SMMC-7721(人肝癌细胞)、A549(人肺腺癌细胞)、Eca-109(人食管癌细胞)、KB(人口腔上皮癌细胞)四种肿瘤细胞,通过将衍生物与母体的抗肿瘤活性的比较寻找KTDA和AKBA结构与活性存在的构效关系。
