荭草素-2"-O-β-D-半乳糖苷抑制小胶质细胞炎症反应及其机制研究

目的:研究荭草素-2"-O-β-D-半乳糖苷(2"-O-beta-D-galactopyranosylorientin,OGA)在脂多糖(LPS)诱导的小胶质细胞炎症模型中的作用及其作用机制。 方法:首先采用Griess法和ELISA检测从金莲花中提取出的黄酮类化合物对LPS诱导的一氧化氮(NO)和肿瘤坏死因子(TNF-α)影响。实时定量PCR检测iNOS,COX-2,TNF-α,IL-1β基因水平表达变化,western blot检测iNOS和COX-2蛋白水平表达变化。细胞免疫荧光检测NF-κB核转位变化,westen blot检测IκB-α降解和MAPKs磷酸化蛋白表达情况。在小胶质细胞/神经元细胞共培养系统检测其神经保护作用。 结果:从金莲花中提取的三种黄酮类化合物均能抑制LPS诱导的NO和TNF-α释放,其中荭草素-2"-O-β-D-半乳糖苷的抑制作用最为显著。荭草素-2"-O-β-D-半乳糖苷显著抑制iNOS,COX-2,TNF-α和IL-1β在基因水平表达,并能抑制iNOS和COX-2蛋白水平表达。荭草素-2"-O-β-D-半乳糖苷能够抑制IκB-α降解,NF-κBp65亚基的核转位以及ERK1/2磷酸化。荭草素-2"-O-β-D-半乳糖苷对神经元细胞具有保护作用。 结论:荭草素-2"-O-β-D-半乳糖苷能够抑制小胶质细胞炎症反应和产生神经保护作用。其主要机制是抑制NF-κB核转位以及ERK1/2磷酸化。
