黄山花菇多糖的化学成分及理化性质研究

黄山花菇是生长在安徽黄山地区的一种大型珍稀食用真菌,以个大、营养丰富及味道鲜美著称。本文采用现代提取分离技术、结构表征手段、药理学实验、流变学方法及结合高分子物理理论知识对黄山花菇中主要活性化学成分多糖进行了系统的研究,获得的研究结果如下:(1)从黄山花菇中分离纯化出4种均一中性多糖组分FMP1、FMP2-1、FMP3-1和FMPS,分子量分别为9.21×106Da、8.4x 106Da、5.74×105 Da和7.2x105 Da。单糖组成分析表明FMP1由甘露糖、葡萄糖和半乳糖组成,摩尔比率为1.00:9.33:1.02;FMP2-1和FMPS由葡萄糖组成;FMP3-1由甘露糖、葡萄糖和半乳糖组成,摩尔比率为1.00:9.23:0.5。结构分析结果表明:FMP1的主链由(1→4)-β-D-Glcp和(1→4)-α-D-Manp组成,在(1→4)-β-D-Glcp的O-2和O-3位,(1→4)-α-D-Manp的O-3位连接有支链(1→3)-β-D-Glcp、(1→6)-β-D-Glcp、 T-α-D-Glcp和T-β-D-Galp; FMP2-1的主链由(1→4)-α-D-Glcp和(1→6)-β-D-Glcp组成,在(1→4)-α-D-Glcp的O-6位连有支链T-α-D-Glcp; FMP3-1的主链由(1→4)-β-D-Glcp、(1→6)-β-D-Glcp和(1→6)-β-D-Manp组成,在(1→6)-β-D-Glcp和(1→6)-β-D-Manp的O-3位连有支链T-β-D-Glcp和T-β-D-Galp; FMPS主链由(1→3)-β-D-Glcp组成,在主链O-6的位置连有支链T-β-D-Glcp,高级结构研究表明FMPS为三螺旋结构。(2)流变学分析表明多糖FMPS溶液为假塑性流体,储能模量G'和损耗模量G"对溶液的浓度和温度有很强的依赖性;指数n(G'~ωn)和损耗角正切tan δ对于浓度和温度具有依赖性;FMPS溶液在15℃的凝胶点(Cgel)为1.16×10-2g/mL,在浓度为1.4×10-2g/mL时的凝胶点(Tgel)为20.6℃; FMPS凝胶不具有时间依赖性且具有热可逆性。(3)硫酸酯化分子修饰结果表明多糖FMPS的硫酸酯化最佳条件为反应温度100℃,氯磺酸与吡啶的比例1:4,反应时间4 h,在此条件下多糖FMPS的硫酸基取代度可达到2.08。(4)生物活性检测结果表明多糖FMP1、FMP2-1和FMP3-1对巨噬细胞RAW264.7的增殖均有促进作用,能增强巨噬细胞的吞噬活性,能刺激巨噬细胞产生NO,且支链的缺失会使其活性减弱;多糖FMPS对于DPPH自由基和羟基自由基有很强的清除能力,当浓度为5mg/mL时,清除率分别为79.46%和74.18%,同时它还具有一定的Fe2+螯合能力和ABTS自由基清除能力;FMPS-S对肝癌细胞HepG2具有不同的抑制活性,其中FMPS-S2在各个浓度下均有最好的肿瘤抑制活性,最高抑制率达42.30%。研究结果对黄山花菇及黄山花菇多糖的开发利用具有一定的指导意义。
