百合知母汤抗抑郁作用的理论与实验研究

摘要
百合病见于张仲景的《金匮要略》,与情志失调关系密切。其临床表现与抑郁症的诊断标准有较明显的相似之处。百合知母汤为治疗百合病的方药之一,由百合和知母两味药组成。本课题系统研究了百合知母汤的化学成分,鉴定22个化合物,其中1个为新化合物,其它化合物均为首次从百合知母汤中分离得到。针对其富含的甾体皂苷,确定了总皂苷的提取和纯化工艺,并建立了皂苷的含量测定方法。采用小鼠尾悬挂、大鼠强迫游泳和慢性应激模型,对百合知母汤和总皂苷的抗抑郁作用及作用机制进行了研究,结果提示,百合知母总皂苷是百合知母汤抗抑郁作用的活性成分之一;百合知母总皂苷抗抑郁作用很有可能与其增加脑内神经递质、逆转HPA轴功能亢进等有关。本课题阐明了经典方百合知母汤抗抑郁的物质基础,同时对其抗抑郁有效部位总皂苷的作用机制进行了初步探讨,为抗抑郁药物的研究奠定了科学基础。
