白术内酯Ⅱ促进大肠癌Lovo细胞凋亡及对PARP1和caspase-3表达的影响

摘要
目的:筛选可有效作用于大肠癌Lovo细胞的中药单体,研究中药单体对间隙连接蛋白Cx43表达的影响,探讨白术内酯Ⅱ对Lovo细胞增殖及凋亡的影响及其机制。方法:体外培养大肠癌Lovo细胞,采用不同浓度的几种中药单体黄芪甲苷、薯蓣皂苷元、黄芩苷、白术内酯Ⅱ分别作用于细胞。噻唑蓝法(MTT法)检测每种中药单体对Lovo细胞增殖的影响;蛋白质印迹法(WesternBlot)检测每种中药单体对Lovo细胞间隙连接蛋白43(Cx43)表达的影响;流式细胞术(FCM)检测白术内酯Ⅱ对Lovo细胞凋亡的影响;蛋白质印迹法(Western Blot)检测白术内酯Ⅱ对Lovo细胞PARP1、caspase-3表达的影响。结果:(1)不同浓度黄芪甲苷、薯蓣皂苷元对大肠癌Lovo细胞增殖无影响;黄芩苷可促进Lovo细胞的增殖,但与浓度无关;不同浓度白术内酯Ⅱ可抑制Lovo细胞增殖,随着浓度的升高,对Lovo细胞增殖的抑制作用越强,细胞存活率逐渐下降,当浓度超过300μg·mL-1时,其抑制率接近50%。(2)不同浓度的四种中药单体黄芪甲苷,薯蓣皂苷元,黄芩苷,白术内酯Ⅱ对大肠癌Lovo细胞间隙连接蛋白43(Cx43)的表达均无影响。(3)低剂量(50μg·mL-1)、中剂量(150μg·mL-1)、高剂量(300μg·mL-1)白术内酯Ⅱ均可促进大肠癌Lovo细胞凋亡,凋亡率与药物浓度相关,差异有显著统计学意义(P<0.01)。(4)白术内酯Ⅱ 100μg· mL-1时使PARP1和caspase-3开始发生剪切,随着药物浓度的增加,其剪切作用明显增强,提示白术内酯Ⅱ可明显改变PARP1和caspase-3的表达,可能通过PARP1和caspase-3途径促进Lovo细胞的凋亡。结论:四种中药单体中只有白术内酯Ⅱ可明显抑制大肠癌Lovo细胞的生长增殖,且呈浓度依赖性。白术内酯Ⅱ还可调节PARP1、caspase-3的表达,促进细胞凋亡。白术内酯Ⅱ能显著抑制Lovo细胞的增殖并诱导其发生凋亡,其分子机制与调节PARP1、caspase-3的表达相关。
