中药丹参和蒲黄的药代动力学研究

药物代谢研究在药物研究与开发上占举足轻重的地位,中药活性成分的代谢研究近几年也越来越引起人们的重视。本文采用CE和HPLC分析手段研究了中药丹参和蒲黄的药代动力学,取得了如下成果: 一、建立兔血浆中丹参素的MEKC分析方法。采用高氯酸沉淀蛋白,乙酸乙酯萃取兔血浆中的丹参素,在MEKC优化条件下,对样品进行分析。该方法的线性范围为0.4~400μg/mL,LOD为0.08μg/mL,精密度的RSD为1.1~5.8%,平均回收率为84.95~104.20%。结果表明,所建立的MEKC方法线性范围宽,灵敏度高,准确性好,适合于丹参素的药代动力学研究。 二、以丹参水提液给兔灌胃,采用MEKC方法研究丹参素在兔体内的药代动力学。药时曲线和药代动力学参数表明:血中丹参素浓度的经时变化符合二室开放模型,兔灌胃丹参水提液后46.9 min达到最大血药浓度,h1/2α为46.9 min,t1/2β为69.3min。 三、建立了兔血浆中槲皮素、山奈素和异鼠李素的RP-HPLC同时分析法。以三氯乙酸为蛋白沉淀剂,水饱和正丁醇与乙酸乙酯的混和溶剂为最佳萃取剂。采用的色谱柱为C18(4.6 mm×150mm,5μm),流动相为0.1%磷酸水溶液:乙腈=73:27。槲皮素、山奈素和异鼠李素的线性范围均为0.05~2.5μg/mL;LOD分别为0.03μg/mL、0.02μg/mL和0.02μg/mL;精密度和准确度均在±15%以内。该分析方法灵敏、准确,适合于蒲黄中黄酮类化合物的药代动力学研究。 四、健康新西兰大白兔单剂量灌胃蒲黄的醇提水解液,以RP-HPLC法同时测定给药后不同时间点槲皮素、山奈素和异鼠李素的血药浓度,用DAS程序计算药动学参数。结果表明:三个指标成分的药动学过程均符合二室开放模型。槲皮素、山奈素和异鼠李素的Tmax分别为32.5,35.0和35.0 min;t1/2α分别为41.67,16.16和29.73 min;V1/F分别为14.18,9.48和84.01 L/kg。 五、针对蒲黄中槲皮素、山奈素和异鼠李素三种黄酮类成分生物利用度低、溶解性差的问题,拟用环糊精包合作用提高其生物利用度。采用淌度移动法研究这三种黄酮类化合物与环糊精的包合常数及包合反应的热力学参数,初步研究其包合过程的机理。
