多巴胺受体D2在纹状体及中脑内的表达

摘要
目的:利用组织芯片及免疫组织化学技术研究多巴胺受体D2在纹状体及中脑内的表达情况,比较两个脑区的多巴胺受体D2表达强度的差异是否具有统计学意义,分忻多巴胺受体D2的分布与解剖结构的关系,为药物成瘾的形成机制提供实验支持。 方法:收集尸检无肉眼病变的人脑标本5例,使用定位取材仪将整个脑组织切成5*5*5mm的小块,并记录每小块脑组织所对应的坐标值,经常规固定、脱水、透明、浸蜡及包埋制作成石蜡蜡块,然后采用组织芯片制备仪并制作成组织芯片。用兔抗人多巴胺受体D2多克隆抗体进行免疫组织化学染色,检测多巴胺受体D2在纹状体及中脑内的表达及分布情况。并运用sPss软件对所得数据进行统计学分忻。 结果:1、多巴胺受体D2在纹状体与皮质、白质、海马表达强度比较,各脑区的表达强度不相同(P<0.05),表达强度的差异有统计学意义。表达强度由高到低依次为:纹状体、海马、皮质、白质。 2、多巴胺受体D2在中脑与皮质、白质、海马表达强度比较,各脑区的表达强度不相同(P<0.05),表达强度的差异有统计学意义。表达强度由高到低依次为:中脑、海马、皮质、白质。 3、纹状体及中脑内的多巴胺受体D2表达强度的差异无统计学意义(P>O.05)。 结论:多巴胺受体D2在纹状体及中脑呈高表达。
