白头翁皂苷A诱导多发性骨髓瘤凋亡及分化的初步研究

摘要
【目的】1.研究白头翁皂苷A对多发性骨髓瘤细胞株U266的抗肿瘤作用。2.观察白头翁皂苷A作用下U266中Bcl-2、cyclin B1等蛋白的变化,部分揭示白头翁皂苷A诱导U266凋亡的作用机制。3.探讨白头翁皂苷A是否能诱导U266细胞的分化。4.观察白头翁皂苷A对多发性骨髓瘤原代细胞的影响。【方法】1.不同浓度的白头翁皂苷A处理人骨髓瘤细胞U266细胞株后,用CCK-8检测法检测细胞增长抑制率,取IC50浓度值处理细胞,在不同时间用同样方法检测细胞生长率,再利用流式检测细胞的周期分布及凋亡率。2.不同浓度的白头翁皂苷A处理U266细胞后,采用Western blot方法检测对细胞抗凋亡蛋白Bcl-2及周期蛋白cyclin B1的影响。3.不同浓度的白头翁皂苷A处理U266细胞后,采用刘氏染色法观察细胞形态变化及流式细胞术检测细胞表面CD49e的阳性表达率,并用ELISA试剂盒分别检测细胞培养上清液中κ轻链蛋白、λ轻链蛋白的浓度。4.通过流式细胞术检测多发性骨髓瘤原代细胞膜表面相关抗原的表达来评价白头翁皂苷A对多发性骨髓瘤原代细胞的影响。【结果】1.白头翁皂苷A对多发性骨髓瘤细胞的抗肿瘤作用在一定范围内U266细胞的增长抑制率随白头翁皂苷A的浓度增高而增高,取IC50(8ng/ul)作用于U266细胞,与对照组相比抑制作用随时间的增加更突出(呈时间依赖性),并且处理24小时后,一部分细胞增长阻滞在了G2期;低浓度白头翁皂苷A对U266细胞的凋亡作用已很明显。2.白头翁皂苷A对多发性骨髓瘤促凋亡可能的作用机制在一定浓度的白头翁皂苷A作用24小时后,抗凋亡蛋白Bcl-2随药物浓度增加而表达下调,G2期调控蛋白随浓度增加而表达增强。3.白头翁皂苷A对多发性骨髓瘤细胞的分化作用不同浓度的白头翁皂苷A可引起U266细胞CD49e的表达上调,并且在形态上趋向更成熟的浆细胞形态,同时细胞培养上清液中κ轻链蛋白、λ轻链蛋白的浓度增加。4.白头翁皂苷A对多发性骨髓瘤原代细胞的影响白头翁皂苷A能够上调多发性骨髓瘤原代细胞表面分化抗原CD49e的表达,使细胞更趋向成熟。【结论】1.白头翁皂苷A对多发性骨髓瘤细胞有促凋亡的作用,且呈时间-剂量依赖性。2.白头翁皂苷A可能是通过Bcl-2、cyclin-B1等蛋白起到促凋亡的作用。3.小剂量白头翁皂苷A可诱导骨髓瘤细胞株进一步的分化,并使其分泌蛋白增加。4.白头翁皂苷A可诱导多发性骨髓瘤原代细胞更趋成熟化。
