基于网络药理学的黄精赞育胶囊治疗少弱精子症作用机制的研究

摘要：

[研究目的]探究黄精赞育胶囊治疗少弱精子症的多成分、多靶点、多途径的作用机制,建立其“化合物-靶点-通路-疾病”的网络药理研究模式。[研究方法]针对黄精赞育胶囊中7个入血成分,使用ChemDraw软件绘制其化学结构,并导入PharmMapper服务器,进行潜在靶点的预测分析,快速检索而获得化合物的靶点信息,并以匹配值(FIT)作降序排列将结果返回,取匹配值排前10位的蛋白作为该化合物的重要靶标蛋白。将蛋白靶点的名称输入UniProt数据库,查询其作用功能,并得到蛋白靶点的官方命名。同时,在UniProt数据库获取这些靶点所对应的基因名称。再通过Molecule Annotation System(MAS 3.0)服务器导入蛋白靶点的名称以获取相关通路,并利用Kyoto Encyclopedia of Genes and Genomes(KEGG)数据库对获取的通路进行分析,最后利用Cytoscape软件绘制黄精赞育胶囊的“化合物-靶点-通路-疾病”网络图。[研究结果与结论]网络分析表明,7个入血成分的作用蛋白靶点约为49个,共涉及通路72条,其中22个靶点蛋白和30条通路可能与少弱精子症相关。黄精赞育胶囊可能通过作用于和激素调节、细胞凋亡、细胞结构、代谢、炎症及免疫相关的蛋白靶点及通路,起到治疗少弱精子症的作用。
