摘要
Original strain,Corynebacteriun glutamicum S9114,was mutated by γ-radiation and DES.Then a strain SH-44,producing Glutamine,was obtained.Fermentatin was conducted in 500mL flask,each containing 30mL of medium consisted of 18% glucose,4% NH -4Cl,1.4% urea,0.5% cornsteepliquor,0.05%K -2HPO,0.05%KH -2PO,0.05%MgSO -4,0.0005%Fe 2+,0.0025% Mn 2+,0.0005% Zn 2+ and 5% CaCO 3.The optima culture condition were as below:initial pH7.0,34℃,shaking speed 300r/min,and cultivation time 72 d.Under optimum condition,SH-44 was able to accumulate an average of 55.3 mg/mL and the highest of 55.8mg/ml of L-glutamine.
