基于网络药理学探讨桂枝芍药知母汤治疗痛风的作用机制

摘要
目的基于网络药理学探讨桂枝芍药知母汤治疗痛风的作用机制。方法结合中药系统药理学数据库与分析平台和文献筛选桂枝芍药知母汤的活性成分和对应靶点。借助GeneCards、OMIM、DisGeNET及TTD数据库检索痛风相关靶点,并将药物与疾病靶点取交集。运用Cytoscape 3.7.2软件构建疾病-药物-靶点-成分网络和蛋白质-蛋白质相互作用网络,借助R 4.0.3软件进行GO富集分析和KEGG通路富集分析。结果预测桂枝芍药知母汤的活性成分125个,药物-疾病共同靶点60个。GO功能富集分析主要涉及磷酸酶结合、金属内肽酶活性、MAPK活性、转录因子结合、氧化还原酶活性等分子功能,KEGG富集分析显示TNF信号通路、TLR信号通路、IL-17信号通路、Th17细胞分化、CLR信号通路、NF-κB信号通路等与痛风密切相关。结论桂枝芍药知母汤在治疗痛风的炎症反应、尿酸代谢紊乱和骨破坏等多个环节中发挥显著作用,为今后临床试验和实验研究提供理论依据。
