内质网应激与炎症对2型糖尿病胰岛β细胞凋亡的影响研究进展

摘要
2型糖尿病(type 2diabetes mellitus,T2DM)是一个以高血糖症为特征的复杂的代谢紊乱过程,其病因是胰岛抵抗和胰岛β细胞功能障碍。越来越多的试验表明,内质网应激(ERS)和炎症反应是导致胰岛β细胞凋亡而引发T2DM的重要原因。胰岛细胞具有发达的内质网,对ERS非常敏感。ERS时,内质网通过未折叠蛋白反应(UPR)调节应激。但持续的应激使UPR反应失调,最终导致胰岛β细胞大量凋亡。另外,炎症反应也可通过UPR反应的介导使胰岛β细胞凋亡。因此,以减轻应激和炎症为目标的治疗手段对于糖尿病治疗有着良好的前景。论文讨论了ERS和炎症过程对于T2DM胰岛β细胞凋亡的影响机制。
