黄芪甲苷对脑缺血小鼠外周免疫抑制作用的影响

摘要
目的观察黄芪甲苷对脑缺血小鼠外周免疫抑制作用的影响,探讨其保护脑缺血损伤的作用机制。方法实验小鼠随机分为假手术组、模型组和给药组,采用大脑中动脉栓塞法制备脑缺血模型,给药组腹腔注射黄芪甲苷溶液。TTC法检测脑梗死体积,神经功能缺损评分评价小鼠神经功能,观察小鼠脾脏质量变化,流式细胞术检测脾脏和外周血中自然杀伤(NK)细胞比例及NKG2D的表达。结果与假手术组比较,模型组小鼠脑梗死体积增加(P<0.001),神经功能缺损评分明显升高(P<0.001),脾脏质量明显减轻(P<0.001),脾脏和外周血中NK细胞比例明显减少(P<0.05,P<0.001),NKG2D表达明显降低(P<0.001);与模型组比较,给药组小鼠脑梗死体积明显减少(P<0.001),神经功能缺损评分明显降低(P<0.001),脾脏质量明显增加(P<0.05),脾脏和外周血中NK细胞比例明显增加(P<0.05),NKG2D表达明显升高(P<0.001)。结论黄芪甲苷对脑缺血损伤小鼠有明显的保护作用,其机制可能与改善外周免疫抑制作用有关。
