发酵冬虫夏草菌丝体中虫草多糖含量的检测及结构鉴定

冬虫夏草是一种中国传统的真菌药材,水溶性虫草多糖作为冬虫夏草菌丝体中的一种重要活性成分,有其独特的生物学活性,利用超声提取法可以分离得到虫草多糖。本研究通过采用苯酚-硫酸法测得中国被毛孢中多糖含量为3.848%。经由热水提取、乙醇沉淀后,通过DEAE-Sephadex A-25离子交换柱对中国被毛孢胞外多糖进行了分级,Sephacryl S-100 HR层析柱进一步分离多糖,得到中性多糖HSP-1以及酸性多糖HSP-2,其分子量分别为3.5伊103k D和3.4伊103k D。结合酸水解、乙酰化、GC-MS、FT-IR、1H NMR以及13C NMR等方法对两个组分的结构进行表征。GC-MS检测确定HSP-1和HSP-2的单糖组成为甘露糖、半乳糖和葡萄糖,摩尔比分别为0.34:0.38:1及0.28:0.24:1。红外光谱和核磁共振的结果表明,HSP-1和HSP-2主要由β-糖苷键连接,HSP-1含有少量α-糖苷键。
