泛素连接酶Cbl-b调控p38MAPK在胰岛素与硒协同抑制糖尿病心肌病大鼠心肌细胞凋亡中的作用

摘要
目的初步探讨胰岛素和硒协同抑制糖尿病心肌病大鼠心肌细胞凋亡的机制。方法 SD大鼠50只,随机分为空白对照组(Control组)、糖尿病心肌病模型组(DCM组)、糖尿病心肌病+胰岛素组(DCM+In组)、糖尿病心肌病+硒组(DCM+Se组)、糖尿病心肌病+胰岛素+硒组(DCM+In+Se组)。TUNEL法观察心肌细胞凋亡;Western blot观察凋亡相关蛋白Bcl-2、caspase-3和PARP剪切片段的变化,同时观察Cbl-b和p38MAPK表达变化;免疫沉淀法检测Cbl-b与p38MAPK、Ku70与Bax之间的相互作用。结果胰岛素与硒协同抑制心肌细胞凋亡(P<0.01);胰岛素联合硒协同,增加Cbl-b的表达,降低p38MAPK表达(P<0.01);两药联合协同增加Cbl-b与p38MAPK、Ku70与Bax之间的相互作用(P<0.01)。结论胰岛素与硒通过调控Cbl-b,抑制p38MAPK而阻止Bax转位来协同抑制心肌细胞凋亡。
