Capillary zone electrophoresis as a method for identification of golden kinglip (Genypterus blacodes) species during frozen storage

Capillary zone electrophoresis (CZE) was used to study the effect of frozen storage on the electrophoretic patterns of sarcoplasmic proteins (SAR) extracted from fillets of golden kinglip (Genypterus blacodes) species using fresh species as a reference. CZE was developed using phosphoric acid at pH 1.5 and an uncoated silica capillary (50 mum i.d. x 75 cm). This condition provided an optimum separation of fish SAR in less than 22 min. Protein profiles were reproducible among individuals belonging to the same species, There was less variation in relative migration time than in peak area percentage for each frozen temperature and storage time assayed. Profiles were independent of fish freshness degree: frozen fish showed the same protein profile as their fresh counterpart. CZE allows correct identification of frozen golden kinglip, fish species stored at -18 or -30 degreesC from 0 to 180 days. (C) 2002 Elsevier Science Ltd. All rights reserved.
