摘要
Traditionally, the use of Sanger sequencing was the one used for DNA sequencing, but the need to look for a substitute was urged due to its failure to meet the increasing demand of technology's high costs and low throughput. The 454 Sequencing came into rescue as it has the highest gains in throughput and sequencing speed. This system is a highly miniaturized sequencing system that is based on pyrophosphate sequencing technology. The system has better accuracy, sensitivity, read length, has higher speed, throughput and lower cost than those provided by Sanger-based technologies. Furthermore, included in the study are the theory and practice of traditional pyrosequencing, modifications to standard pyrosequencing, and finally, the right use of 454 sequencing system.
