Supplementing a medium with non-essential amino acids results in an increased membrane stability during the death phase of a batch hybridoma culture (determined by Trypan Blue). The effect can only be seen when the amino acids are added at the beginning of the culture. This then leads to a different metabolism resulting in a decreased lactate concentration and an increased antibody secretion during the death phase. Surprisingly the addition of the amino acids has no effect on the determined viability (acridine orange and propidium iodine staining).
