A review is given on the methodologies for generating a variety of solution-phase combinatorial libraries, including, parallel single-compound libraries, indexed and mixture combinatorial libraries, complex libraries generated from unsymmetrical and constrained symmetrical scaffolds, and libraries produced by chemoenzymatic and multicomponent reaction (MCR) approaches up to the beginning of 1999. Biological activities generated by solution-base methodologies and deconvolution processes of related libraries are summarized. Further, the purification and characterization/confirmation of solution-phase combinatorial libraries are also highlighted.
