微生物转化阿魏酸生成香草醛的研究

摘要
本论文对黑曲霉(Aspergillus niger)CGMCC0774和朱红密孔菌(Pycnoporus cinnabarinus)CGMCC1115联合转化米糠油脚中的阿魏酸生产香草醛进行研究。 用米糠油脚水解提取的阿魏酸作为生产香草醛的底物。通过60Co诱变黑曲霉(A. niger)SW-33,经过初筛、复筛、再复筛得到一株可耐较高底物浓度的突变株黑曲霉(A. niger)CGMCC0774,三次平均摇瓶转化得到香草酸浓度为2.481 g/L,摩尔转化率为71.62%。研究黑曲霉CGMCC0774转化阿魏酸生成香草酸的最适条件为:转化温度为37℃,转速150 r/min,底物阿魏酸浓度为4 g/L,pH4.0-4.5,摇瓶装液量为140 mL/250 mL。进行黑曲霉CGMCC0774转化阿魏酸生成香草酸15 L和25 L发酵罐放大试验,通过流加底物得到香草酸2.24 g/L,摩尔转化率为64.6%。 研究从阿魏酸到香草醛的两步串联转化工艺,黑曲霉转化阿魏酸生成香草酸的转化液过滤除去菌体,调pH5.0,补加5 g/L葡萄糖,灭菌,用培养好的朱红密孔菌游离菌体转化,产香草醛0.63 g/L。研究朱红密孔菌(P. cinnabarinus)CGMCC1115反应分离耦合转化,当香草酸浓度为4 g/L和树脂加量为20 g时,香草醛浓度为2.20 g/L,摩尔转化率为60.69%。 将纯化后得到的本课题研制香草醛结晶样品进行13C同位素测定,其碳13C/12C比率明显不同于愈创木酚原料化学合成的香草醛,与Rhovanil? Natural(天然等同)相近。 尝试对朱红密孔菌细胞固定化的初步研究,转化72 h产香草醛0.499 g/L。固定化细胞反复利用四次,香草醛浓度明显下降,朱红密孔菌的固定化有待于进一步研究。
