川陈皮素抑制IL-21/IL-21R介导类风湿关节炎的炎症反应研究

摘要
第一部分:川陈皮素抑制IL-21/IL-21R介导类风湿关节炎的炎症反应体外研究目的:充分探讨川陈皮素靶向IL-21调控类风湿关节炎免疫应答过程,确定其与IL-21/IL-21R及JAK1/STAT3信号通路在调控过程中的介导关系,确定川陈皮素在类风湿关节炎发生、发展过程中的价值性,为后期临床治疗类风湿关节炎提供研究方向。材料与方法:我们使用的是RASFs细胞系,即MH7A细胞,它来自RA患者,被认为是RA的主要效应细胞。MH7A细胞系使用含10%胎牛血清的1640完全培养基于37℃、5%CO2、100%的恒温培养箱中培养,具体分组如下:Control组(不做处理);IL-21组(50 ng/ml IL-21处理MH7A细胞);IL-21+Nobiletin 25μM组(50ng/ml IL-21+25μmol/L Nobiletin处理MH7A细胞);IL-21+Nobiletin 50μM组(50ng/ml IL-21+50μmol/L Nobiletin处理MH7A细胞);MH7A细胞干预结束后,继续培养48小时后进行相关项目分析:细胞因子分析:ELISA检测各组细胞中IL-6、TNF-α、HMGB1、MMP-3、MMP-13表达浓度分析;各组MH7A细胞ROS(活性氧)检测;各组MH7A细胞4-HNE(4-羟基壬烯醛)检测;RT-PCR检测IL-21R、IL-6、TNF-α、MMP-3、MMP-13表达浓度分析;Western-blot检测IL-21R及JAK1/STAT3信号通路关键蛋白表达分析;统计数据,给出结论。结果:ELISA化学检测:IL-6、TNF-α:各组MH7A细胞中的表达趋势为Control0.05,差异不具有统计学意义;在其磷酸的表达过程中,各组MH7A细胞中的表达趋势为Control0.05,差异不具有统计学意义;在其磷酸的表达过程中,表达趋势为Control<IL-21+Nobiletin 50μM<IL-21+Nobiletin 25μM<IL-21,P<0.05,差异具有统计学意义。结论:(1)川陈皮素可以较好的改善模型组大鼠关节炎效应,明显改善大鼠的精神状态,降低大鼠的类风湿关节炎症的程度及关节指数,为后续的研究提供帮助。(2)与体外研究结果一致,川陈皮素改善大鼠类风湿关节炎症的作用主要是通过靶向机体IL-21R浓度变化进而调控JAK1/STAT3的活性状态,充分降低白细胞介素、肿瘤坏死因子-α、高迁移率蛋白表达,逐渐恢复关节软骨基质合成及分解的平衡状态,加速类风湿关节炎疾病的康复过程,达到对类风湿关节炎的治疗效果,给予临床一定启发。
