INVESTIGATION OF THE CONJUGASE TREATMENT PROCEDURE IN THE MICROBIOLOGICAL ASSAY OF FOLATE

摘要
Microbiological assays have been conducted to determine the growth responses of Lactobacillus casei to pteroyl di-, tri-, tetra-, hexa-, and hepta- glutamates relative to the monoglutamate (folic acid). Deconjugation using human blood plasma conjugate at acidic pH (4.5) results in a substantial degradation of folates while between pH 6.0 and 7.0, optimum results could be obtained using a slight excess of plasma. The end products of conjugase treatment with either plasma or chicken pancreas extract as the source of conjugase depend on, besides the conjugase source, the incubation pH and the inhibitory components present in the incubation mixture.
