当归多糖对小鼠骨髓造血干细胞及基质细胞表面细胞间黏附分子1水平的影响

背景:有研究表明当归多糖有补血活血作用,但其如何改善骨髓造血微环境,促进造血功能成为研究问题热点之一。目的:观察当归多糖对小鼠骨髓造血干细胞及基质细胞表面细胞间黏附分子1水平的影响。方法:建立失血性贫血模型小鼠,随机分为3组,当归多糖低、高剂量组连续腹腔注射等量相应浓度的当归多糖4,6 mg/kg,对照组则同样时间内注射等量的生理盐水,共干预6 d。动态观察小鼠一般情况;全自动血细胞分析仪检测不同时间点小鼠外周血中红细胞的数量;采用磁珠分选和流式细胞术检测骨髓中造血干细胞的数量;MTT法检测骨髓基质细胞的增殖情况;采用流式细胞术检测骨髓基质细胞表面的细胞间黏附分子1的表达。结果与结论:当归多糖对失血性贫血小鼠体质量无明显影响(P>0.05)。当归多糖低、高剂量组外周血红细胞数、骨髓CD34+,Sca-1+细胞百分数、骨髓基质细胞数量及其表面细胞间黏附分子1均明显高于对照组(P<0.05),其中高剂量组效果优于低剂量组。结果说明,当归多糖通过促进骨髓基质细胞增殖,增强细胞间黏附分子1的表达,进而促进造血干细胞增殖并影响其功能活动。
