茅苍术和白芷化学成分研究

摘要
茅苍术Atractylodes lancea(Thunb)DC.(又名南苍术)为菊科(Compositae)苍术属植物,是燥湿、祛风的中药材,应用历史悠久,畅销国内外。茅苍术主要产于江苏、湖北、浙江、安徽,以产于江苏茅山一带质量为佳,因此得名。目前国内外有关茅苍术的化学成分研究多针对其挥发油部分,对非挥发油成分因其难度较大而研究的较少。为了更好的了解、阐明其化学成分组成和药理药效基础,本文对茅苍术的非挥发油化学成分进行了研究。 利用多种现代色谱分离技术和分离分析仪器,从茅苍术新鲜根茎的乙醇提取物中共分离得到了12个化合物,鉴定了其中8个:苍术烯内酯丙(atractylenolidⅢ,CZ-3),β-谷甾醇(β-sitosterol,CZ-4),胡萝卜苷(β-daucosterol,CZ-5),苍术苷A(atractyloside A,CZ-6),3,4,11,14-四羟基愈创-9-烯-11-O-β-D-吡喃葡糖苷(3,4,11,14-tetrahydroxyguai-9-en-11-O-β-D-glucopyranoside,CZ-7),(1S,5R,7R,10R)-裂苍术内酯-11-O-β-D-吡喃葡糖苷[(1S,5R,7R,10R)-secoatractylolactone-11-O-β-D-glucopyranoside,CZ-8)],棕榈酸(Palmitic acid,CZ-9),(1S,4S,5R,7R,10R)-11,14-二羟基愈创-3-酮-11-O-β-D-吡喃葡糖苷[(1S,4S,5R,7R,10R)-11,14-dihydroxyguai-3-one 11-O-β-D-glucopyranoside,CZ-11A)]。其中化合物CZ-7是新化合物。 中药白芷(Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.vat.formosana(Boiss.)Shah et Yuan)为伞形科(Umbeliferae)当归属(Angelica)植物。白芷是常用中药,始载于《神农本草经》,列为中品。但是目前关于白芷的化学成分研究主要集中在中小极性的香豆素类成分方面,关于极性较大的苷类成分研究较少。为了更好的开发利用此种传统中药,明确其化学成分组成和药理药效作用,我们对白芷水溶性化学成分进行了研究。 从白芷乙醇提取物中分离鉴定了7个苷类成分:7-O-β-D-Apiofuranosyl-(1→6)-β-D-Glucopyranosyl-peucedanol(AD-1),(R)-peucedanol-7-O-β-D-Glucopyranoside(AD-2),aesculin(esculin)(AD-3),(3′R)-Hydroxymamesin-4′-O-β-D-Glucopyranoside(AD-4),sec.-O-β-D-Glucopyranosyl-(R)-byakangelicin(AD-5),tert.-O-β-D-Glucopyranosyl-(R)-byakangelicin(AD-6)和胡萝卜苷(β-daucosterol)(AD-7)。其中化合物AD-1和AD-2均首次从当归属中分离得到,化合物AD-3首次从伞形科中分离得到。
